Neutrophil iodination reaction induced by fluoride: implications for degranulation and metabolic activation.

چکیده
Activation of neutrophils by phagocytosis or certain soluble stimuli results in a burst of oxidative metabolism and in degranulation. Fluoride is known to trigger the metabolic burst. but degranulation has not been reported. lodination of protein by F -activated human PMN was demonstrated by measuring TCA-precipitable 1251. lodination was dependent on H20,. since it was inhibited by catalase. enhanced by superoxide dismutase. and absent in PMN from patients with chronic granulomatous disease. Dependence on myeloperoxidase was indicated by an inhibitory effect of azide and cyanide and by the absence of iodination in myeloperoxidase-deficient PMN. Soluble extracellular protein was the predominant substrate for iodination. suggesting secretion of myeloperoxidase and H,O,. Maximum iodination occurred after a 60-mm exposure of PMN to 20-30 mM fluoride. At greater than 30 mM F . there

Cytochemical demonstration of hydrogen peroxide in polymorphonuclear leukocyte phagosomes
Phagocytosis by polymorphonuclear leukocytes (PMN) is accompanied by specific morphological and metabolic events which may result in the killing of internalized micro-organism. Hydrogen peroxide is produced in increased amounts during phagocytosis (17) and in combination with myeloperoxidase and halide ions constitute a potent, microbicidal mechanism (8,9,11). There can be direct iodination of ...
متن کاملRelease of 5'-guanosine monophosphate and adenine by Brucella abortus and their role in the intracellular survival of the bacteria.
Brucella abortus releases two components (fractions 3b and 10) that inhibit the myeloperoxidase-H2O2-halide antibacterial system of bovine neutrophils by inhibiting degranulation. Fractions 3b and 10 were analyzed by high-performance liquid chromatography and thin-layer chromotography and compared with nucleotide and base standards. These investigations indicated that fraction 3b coeluted and c...
متن کاملSpecific and azurophilic granules from rabbit polymorphonuclear leukocytes. II. Cell surface localization of granule membrane and content proteins before and after degranulation
The compositional relationship between the cell surface of rabbit polymorphonuclear leukocytes (PMNs) and the membranes of PMN cytoplasmic granules has been investigated. Heterophilic PMNs obtained from peritoneal exudates contained 13 cell surface polypeptides ranging in molecular weight from 220,000 to 12,000 daltons as determined by lactoperoxidase-catalyzed protein iodination and gel electr...
متن کاملMetabolic activity of phagocytosing granulocytes in chronic granulocytic leukemia: ultrastructural observation of a degranulation defect.
The functional capacities of granulocytes in patients with chronic granulocytic leukemia are still a subject of controversy, probably due to the heterogeneity of the abnormalities observed from patient to patient. For a better definition of these abnormalities, 14 patients with untreated chronic granulocytic leukemia were studied. The patients were divided into three groups on the basis of the ...
متن کاملFilifactor alocis Promotes Neutrophil Degranulation and Chemotactic Activity.
Filifactor alocis is a recently recognized periodontal pathogen; however, little is known regarding its interactions with the immune system. As the first-responder phagocytic cells, neutrophils are recruited in large numbers to the periodontal pocket, where they play a crucial role in the innate defense of the periodontium. Thus, in order to colonize, successful periodontal pathogens must devis...
